A. HEK 293T cells were transfected with 0.5 μg of PURA reporter plasmid, and 4 μg of the expression plasmids pEGFP-C1 (hIRF-1, hIRF-3, hIRF-5, hIRF-7, or hIRF-9) or the control plasmid pEGFP-C1. At twenty-four hours post-transfection cells were viewed by fluorescent microscopy for the expression of GFP. At forty-eight hours post-transfection cells were lysed and analyzed for promoter expression as described in Methods.
